Discovery of a neuroprotective natural product by high throughput screening

Abstract
Glutamate induced oxidative stress is major cause of neuronal cell death. Oxidative stress increases cellular Ca2+ concentration which is upstream of reactive oxygen species (ROS) mediated apoptosis, and MAPK activation. Therefore, antioxidant spices can be utilized as neuroprotective materials against oxidative stress induced neuronal cell death. Natural products contain many anti-oxidant molecules including flavonoids, polyphenols and etc. High through put screening (HTS) enables the identification of biologically active molecules within a short period. In particular, natural products include many kinds of chemical components that are required to evaluate their activity. The present study used HTS combined with high content screen to identify neuroprotective natural products. We identified three natural products which possess neuroprotective properties using high through put screening from 80 natural product extracts obtain from South Korea. We used glutamate-induced neurotoxicity in HT22 hippocampal neuronal cells to identify neuroprotective natural product extracts. In this screening, Hedyotidis Herba was identified as neuroprotective material with more than 80% cell viability at 16.6 §¶/ml. Salix babylonica L showed 100% cell viability at 50 §¶/ml. Lespedeza cuneate G. Don showed 80% cell viability at 50 §¶/ml. Further research on the mode of action of these natural product extracts is underway.
